Is it appropriate to ask for a different runway than the one assigned by the tower for takeoffs / landings? Do I have to explain why I want a different runway? (winds, closer to parking, etc)

It's never inappropriate to ask ATC for a different clearance that the one issued. There is no guarantee you'll get it, but you may as well ask. No need to explain it to them. Remember, the pilot in command is ultimately responsible for the aircraft. Don't accept a clearance you don't like or can't handle.

If I have a specific clearance need or request, I try to get it in on initial contact or at some point before issuance of a contrary clearance, like if I know they're likely to give me a clearance to 34 right, I say "Requesting runway 34 left if able."

And I respectfully disagree with Jeff. I think you should ask for a clearance modification if you need it, and I think most controllers are happy to oblige if they're able to. Good judgement applies, however. You need to take into account the traffic saturation and controller workload before you ask for amendments or special clearances and be reasonable with your requests.
